SM Buffer is a Tris-HCl–based saline–magnesium buffer used for the preparation, dilution, and storage of bacteriophage suspensions. It is commonly applied in plaque assays with E. coli host strains, phage titering, elution from agar plugs, and maintenance of phage lysates.

This sterile, ready-to-use solution has a near-neutral pH and is compatible with standard bacteriophage workflows, including serial dilutions, plate washes, and resuspension of phage concentrates.

  • 100mM Sodium Chloride
  • 10mM Magnesium Sulfate
  • 0.01% Gelatin
  • 50mM Tris-HCl
  • Bacteriophage Storage and Stabilization: Maintains phage integrity during short- and long-term storage with NaCl, Mg2+, and gelatin.

  • Phage Dilution and Titering (Plaque Assays): Used as a diluent for accurate serial dilutions and plaque-forming unit (PFU) determination.

  • Elution of Phage from Agar: Recovers phage from agar plugs/overlays after plaque isolation.

  • Resuspension of Precipitated Phage: Rehydrates PEG/NaCl-precipitated or pelleted phage for downstream use.

  • Preparation and Transport of Phage Lysates: Serves as a stable medium for handling and transferring phage samples between steps.

  • Host Infection and Adsorption Assays: Supports phage adsorption and infection experiments on hosts such as Escherichia coli, Salmonella enterica, and Pseudomonas aeruginosa.

  • Template Preparation for Phage Nucleic Acids: Provides a compatible buffer for phage DNA/RNA extraction workflows.

  • Phage Library and Amplification Workflows: Used as a consistent diluent during library amplification, pooling, and storage.
