Standard Methods Agar (Plate Count Agar) is a general-purpose medium used for enumeration of aerobic heterotrophic bacteria in water, dairy, and food samples. Its balanced nutrient composition supports the recovery and cultivation of a broad range of mesophilic organisms for colony counting.

Typical use includes preparation of pour plates or spread plates for total aerobic plate counts under standard incubation conditions as described in compendial methods.

  • 0.5% Tryptone
  • 0.25% Yeast Extract
  • 0.1% Cerelose Anhydrous Dextrose (Glucose)
  • 1.5% Agar
  • Aerobic Plate Count (APC): Enumeration of viable aerobic mesophilic bacteria as CFU/g or CFU/mL using pour or spread plate techniques.

  • Heterotrophic Plate Count (HPC): Assessment of general heterotrophic microbial load in potable, process, and recreational water.

  • Food and Dairy Microbial Enumeration: Total viable counts in milk, dairy, meat, produce, and ready-to-eat foods for quality and safety programs.

  • Water and Beverage Quality Testing: Monitoring microbial load in bottled water, juices, and process water to verify process control.

  • Environmental Monitoring and Sanitation Validation: Quantification of surface and equipment swab samples to confirm cleaning effectiveness.

  • Bioburden Testing (Non-Sterile Products): Microbial load determination in cosmetics, nutraceuticals, and raw materials.

  • Shelf-Life and Spoilage Assessment: Tracking total aerobic counts over time to evaluate product stability and spoilage risk.

  • Pour Plate and Spread Plate Methods: Standardized CFU enumeration workflows compatible with 100 mm sterile plates (20 plates/sleeve).

  • Recovery of Stressed Organisms: Resuscitation and enumeration of sublethally injured aerobes following heat or sanitizer exposure.

  • Colony Isolation for Downstream ID: Isolation of mixed aerobic populations (e.g., Bacillus, Pseudomonas, Micrococcus, Staphylococcus) for further identification or testing.
