10X SPG (Sucrose Phosphate Glutamate) Solution, Non-Sterile, is a concentrated buffer used to prepare working-strength SPG for preservation, transport, and storage of viral specimens and fastidious intracellular bacteria such as Chlamydia trachomatis and Rickettsia spp. The 10X format allows convenient dilution to 1X with sterile water or saline according to laboratory protocols.

The phosphate buffer maintains pH stability, sucrose provides osmotic and cryoprotective support, and L-glutamic acid serves as an amino acid stabilizer for sensitive biological materials. As a non-sterile concentrate, it should be sterilized or aseptically filtered after dilution when required for cell culture or clinical workflows.

  • 38mM Potassium Phosphate Monobasic
  • 72mM Potassium Phosphate Dibasic
  • 2.18M Sucrose
  • 49mM L-Glutamic Acid Free Acid
  • Viral and Bacteriophage Stabilization: Stabilizes virions and phage particles during freezing, thawing, and storage (e.g., −80°C or lyophilization) to preserve infectivity.

  • Transport Medium for Obligate Intracellular Bacteria: Maintains viability of clinical and research specimens containing Chlamydia trachomatis, Chlamydia pneumoniae, Rickettsia rickettsii, and Coxiella burnetii.

  • Virus/Phage Titration and Dilution Buffer: Used as a stabilizing diluent for plaque assays and serial dilutions to minimize titer loss.

  • Storage of Chlamydial Elementary Bodies: Preserves infectivity of purified elementary bodies for culture, passage, and quantification assays.

  • Resuspension and Washing Buffer: Protects labile viral particles and sensitive bacteria during pelleting, resuspension, and washing steps.

  • Lyophilization Excipient: Serves as a protective matrix for freeze-drying viral stocks and reference materials.
