Rappaport-Vassiliadis Broth (RVS) is a selective enrichment medium used for the detection of Salmonella from food, water, and environmental samples. The formulation uses elevated magnesium chloride and malachite green to inhibit competing flora, while a phosphate buffer, tryptone, and sodium chloride provide pH control, nitrogen and peptide sources, and osmotic balance.

This medium enables selective enrichment of Salmonella spp. prior to isolation on differential or selective agar.

  • 1.34% Magnesium Chloride
  • 0.72% Sodium Chloride
  • 0.45% Tryptone
  • 0.15% Monopotassium Phosphate
  • 36µg/mL Malachite Green Oxalate
  • Selective Enrichment of Salmonella from Foods: Enriches Salmonella from meat, poultry, eggs, dairy, and produce using malachite green (36 µg/mL) and high MgCl2 (1.34%) for selectivity.

  • Environmental Monitoring: Detects Salmonella in swabs and rinses from food processing environments and equipment surfaces.

  • Water and Beverage Testing: Enriches low-level Salmonella from potable, process, and surface waters, juices, and other beverages.

  • Pharmacopeial/Compendial Testing: Used in USP/EP/JP specified organism testing workflows for Salmonella after non-selective pre-enrichment.

  • Regulatory and Standard Methods: Applied in ISO 6579, FDA BAM, USDA/FSIS, and AOAC workflows for Salmonella detection.

  • MPN and Quantitative Enrichment: Supports Most Probable Number schemes to estimate Salmonella levels in complex samples.

  • Suppression of Competing Flora: Inhibits background organisms (e.g., Proteus, coliforms) via malachite green and elevated salts (NaCl 0.72%, MgCl2 1.34%).

  • Pre-Plating Enrichment: Enrichment prior to isolation on selective agars (e.g., XLD, HE, BS) to recover stressed Salmonella cells.

  • Pre-PCR/Immunoassay Preparation: Generates enriched cultures to improve sensitivity of molecular assays and immunoassays targeting Salmonella.

  • Animal Feed and Ingredients Testing: Enrichment of Salmonella from feed, pet food, and raw ingredients with high selectivity from the RVS formulation.
