10X Phosphate Buffer is a concentrated potassium phosphate buffering solution intended to be diluted 1:10 with water to prepare a 1X buffer. When diluted to 1X, it provides near-neutral buffering capacity suitable for general laboratory use.

Applications include preparation of pH-stable reagents, dilution of samples and standards, washing steps in cell handling, and suspension of bacterial cells such as E. coli in workflows where a potassium phosphate system is preferred.

Directions: Mix 1 volume of 10X solution with 9 volumes of high-purity water to obtain 1X. Verify pH of the working solution and adjust if required for the application. Sterilize by filtration or autoclaving as needed.

  • 170mM Monopotassium Phosphate
  • 720mM Dipotassium Phosphate
  • Polymerase Chain Reaction (PCR): Used in PCR reactions to maintain optimal pH conditions for enzyme activity.

  • Gel Electrophoresis: Serves as a running buffer in SDS-PAGE and native PAGE, providing consistent pH for protein separation.

  • Protein Purification: Utilized in anion exchange chromatography to maintain a stable pH environment during protein elution.

  • Enzyme Assays: Provides a consistent pH for enzymatic reactions, ensuring accurate kinetic measurements.

  • Sample Dilution and Reagent Preparation: Acts as a stable pH buffer for preparing standards, controls, and dilutions.

  • Cell Washing and Resuspension: Maintains physiological pH during washing, pelleting, and resuspending cells.

  • ELISA and Immunoassays: Used as a coating and dilution buffer to preserve antigen–antibody interactions at controlled pH.

  • Spectrophotometric Assays: Stabilizes pH for absorbance-based measurements of proteins, nucleic acids, and metabolites.
