1M Sodium Phosphate Buffer, pH 6.0, is an inorganic buffering solution for controlling pH in biological and chemical procedures. It contains 1 mol/L total phosphate in aqueous solution adjusted to pH 6.0.

Typical uses include sample preparation, dilution and wash steps, chromatographic equilibration and elution systems compatible with phosphate buffers, preparation of calibration standards, and dilution to lower working strengths for assay buffers.

  • 1M Sodium Phosphate
  • Enzyme Assays: Provides a stable pH 6.0 environment for enzymes with acidic optima, enabling reproducible kinetic measurements.

  • Protein Purification: Used as equilibration, wash, and elution buffer for ion-exchange chromatography near pH 6.0 to preserve protein stability.

  • Protein Storage and Dialysis: Maintains protein conformation and activity during storage, buffer exchange, and desalting at mildly acidic pH.

  • HPLC Mobile Phase: Serves as a phosphate buffer component for reproducible retention and peak shape in reversed-phase and ion-exchange HPLC at pH 6.0.

  • Sample Preparation and Dilution: Acts as a 1 M stock to adjust pH and ionic strength when preparing working buffers at or near pH 6.0.

  • Cell and Tissue Homogenization: Buffers extracts during lysis and clarification to protect pH-sensitive biomolecules.

  • Microbiology Media Buffering: Stabilizes culture media at mildly acidic pH for organisms and workflows that require pH 6.0; sterile format supports aseptic applications.

  • pH: 6.0 ± 0.04
