20X PBS with 1% Tween-20 is a concentrated phosphate-buffered saline containing a nonionic surfactant for preparation of a 1X working buffer by 1:20 dilution with water. The 1X solution provides isotonic ionic strength and phosphate buffering suitable for routine laboratory workflows.

When diluted to 1X, the final Tween-20 concentration is 0.05%. The buffer is used for washing steps, membrane-based assays, immunostaining procedures, and sample preparation in applications such as Western blotting and ELISA.

  • 2.7M Sodium Chloride
  • 54mM Potassium Chloride
  • 86mM Sodium Phosphate Dibasic
  • 28mM Potassium Phosphate Monobasic
  • 1.0% Tween-20
  • Western Blot Washing: Common wash buffer for membranes to reduce non-specific binding during antibody probing.

  • ELISA Plate Washing: Standard wash buffer for microplates to minimize background and improve signal-to-noise.

  • Immunohistochemistry/Immunocytochemistry: Wash buffer between antibody incubations to preserve antigen-antibody specificity.

  • Immunofluorescence Microscopy: Gentle detergent-containing washes to reduce background without disrupting cells.

  • Antibody Diluent (with blockers): Base buffer for diluting primary/secondary antibodies when combined with BSA or serum.

  • Flow Cytometry: Cell wash buffer to limit cell clumping and non-specific antibody interactions.

  • In Situ Hybridization: Detergent-containing washes to reduce non-specific probe binding on membranes or tissues.

  • Dot/Slot Blotting: Wash buffer for membrane-based nucleic acid and protein detection assays.

  • General Laboratory Washing: Rinsing cells, beads, and surfaces in protein/DNA workflows to lower non-specific adsorption.

  • Coating/Blocking Workflows: Used between coating and blocking steps to remove unbound material and stabilize assay surfaces.
