Plate Count Agar is a nonselective solid medium used for enumeration of viable aerobic and facultative heterotrophic bacteria in food, water, dairy, and environmental samples by pour plate or spread plate methods.

The nutrient base supplies amino acids, peptides, vitamins, and a readily metabolizable carbon source, while agar provides a solid matrix for colony development and counting. Typical uses include total aerobic plate counts and general microbial load assessments of raw materials, process intermediates, and finished products, accommodating a broad range of organisms such as Escherichia coli and Bacillus subtilis.

  • 0.50% Tryptone
  • 0.25% Yeast Extract
  • 0.10% Glucose
  • 2.00% Agar
  • Total Aerobic Plate Count (APC): Enumeration of viable heterotrophic bacteria in food, dairy, beverages, and water using a nonselective medium containing 0.50% Tryptone, 0.25% Yeast Extract, 0.10% Glucose, and 2.00% Agar.

  • Pour/Spread Plate Quantification: CFU determination for routine quality control and shelf-life studies across raw materials and finished products.

  • Microbial Limit Testing: Nonselective growth support for compendial bioburden assessments in pharmaceuticals, nutraceuticals, and cosmetics.

  • Environmental Monitoring: Enumeration from air, surface, and equipment swabs in production and cleanroom settings.

  • Recovery of Stressed Cells: Facilitation of sublethally injured organism recovery after processing or sanitation stress.

  • Water and Beverage Testing: Routine plate counts in potable water, process water, and beverage manufacturing.

  • Process Verification/HACCP: Monitoring microbial load to validate sanitation efficacy and critical control points.

  • Colony Selection for ID: Growth of representative colonies (e.g., Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Bacillus spp., Lactobacillus spp.) for downstream identification workflows.

  • Mixed Culture Dilution Plating: Enumeration from complex matrices to assess community-level viable counts without selectivity bias.

  • Routine QC of Production Lines: Trending bioburden across batches, lines, and shifts for continuous improvement.
