2M MOPS Buffer, pH 7.4, is a concentrated Good’s buffer stock for preparing near-neutral aqueous buffering systems. MOPS (3-(N-morpholino)propanesulfonic acid) has a pKa of approximately 7.2 at 25 °C, providing buffering capacity in the pH 6.5–7.9 range. The 2 M concentration allows preparation of working solutions at commonly used ranges (e.g., 10–100 mM) by volumetric dilution with water or salt solutions.

This buffer is used in biochemical assays, protein and nucleic acid workflows, electrophoresis systems employing MOPS chemistry, and cell culture media where a zwitterionic, low-metal-binding buffer at physiological pH is needed. The pH is adjusted to 7.4 at 25 °C. Prepare working buffers by diluting the 2 M stock to the desired molarity; for example, a 50 mM MOPS buffer is obtained by a 1:40 dilution.

  • 2M MOPS
  • RNA Agarose Gel Electrophoresis: Serves as the buffering component (e.g., MOPS–formaldehyde) for denaturing RNA gels, maintaining neutral pH and RNA integrity.

  • Protein Purification and Chromatography: Maintains stable pH near 7.4 in ion-exchange, size-exclusion, and affinity workflows.

  • Enzyme Assays: Provides consistent near-neutral pH for enzymes where Tris or phosphate buffers are suboptimal.

  • Cell Culture Media Buffering: Supplements media to stabilize extracellular pH in culture systems.

  • MOPS-Defined Minimal Media: Used to formulate minimal media for growth of Escherichia coli.

  • Nucleic Acid and Protein Sample Buffers: Included in preparation and storage buffers to minimize pH drift and interference with downstream assays.

  • pH: 7.4 ± 0.04
