Modified M9 Minimal Salts Media with 0.4% Maltose is a defined minimal medium formulated for cultivation of maltose-utilizing bacteria, including E. coli, using maltose as the sole carbon source. The minimal salt base provides essential inorganic nutrients for controlled growth conditions.

Thiamine supplies a vitamin cofactor required for central metabolic pathways, and magnesium sulfate provides divalent cations necessary for enzymatic function and ribosomal stability. This medium is suited for studies of carbohydrate utilization, physiological characterization, and maintenance of strains adapted to minimal conditions.

  • 1X M9 Salts
  • 0.4% Maltose
  • 1µg/mL Thiamine
  • 2mM Magnesium Sulfate
  • Defined Culturing of Escherichia coli: Growth of E. coli and related enteric bacteria in a minimal, chemically defined medium using maltose as the sole carbon source.

  • Maltose Utilization and Transport Studies: Selection and characterization of mal operon mutants and assays of maltose/maltodextrin transport systems.

  • Lambda Phage Workflows: Induction of LamB (maltoporin) for efficient bacteriophage λ adsorption, infection, and transduction experiments in E. coli.

  • Recombinant Protein Expression in Minimal Media: Controlled expression in E. coli under defined conditions with maltose to modulate catabolite repression and overflow metabolism.

  • Carbon Source and Catabolite Repression Studies: Comparative growth and regulatory analyses (CRP–cAMP) using maltose versus other sugars.

  • Auxotrophy Testing and Complementation: Phenotyping and selection of metabolic mutants in a minimal background supplemented with thiamine.

  • Antibiotic Selection in Defined Conditions: Plasmid maintenance and selection where complex media components are undesirable.

  • Chemotaxis and Motility Assays: Evaluation of bacterial taxis toward maltose gradients in minimal conditions.

  • Growth Kinetics and Bioprocess Optimization: Determination of doubling times, yield coefficients, and metabolic flux analyses with maltose as the carbon source.
