Minimal M9 Broth with 0.2% Glucose is a defined salts medium formulated for cultivation of E. coli under minimal conditions. Glucose serves as the sole carbon source, thiamine supplies an essential vitamin for thiamine-requiring strains, and magnesium and calcium provide essential divalent cations for cellular processes.

This medium is suitable for experiments requiring controlled, defined nutrient composition, including physiological studies, carbon-source regulation, and maintenance of prototrophic or thiamine-requiring E. coli strains.

  • 1X M9 Salts
  • 0.2% Cerelose Anhydrous Dextrose (Glucose)
  • 0.5µg/mL Thiamine
  • 1mM Magnesium Sulfate
  • 0.1mM Calcium Chloride
  • Defined Cultivation of Escherichia coli: Supports growth under precisely defined conditions with glucose, Mg2+, Ca2+, and thiamine.

  • Metabolic Mutant Selection and Auxotrophy Testing: Differentiates prototrophs from auxotrophs and enables complementation analysis on minimal nutrients.

  • Carbon Utilization and Catabolite Repression Studies: Evaluates glucose metabolism and regulatory responses in a controlled minimal medium.

  • Recombinant Protein Expression in Defined Media: Provides consistent growth and reduced byproducts for expression in E. coli.

  • Quantitative Physiology and Growth Kinetics: Enables reproducible growth curves, yield measurements, and nutrient-limitation studies.

  • Antibiotic Selection and Plasmid Maintenance: Maintains selectable markers with minimal background components that can confound phenotypes.

  • Chemotaxis and Motility Assays: Offers a defined matrix with glucose to assess movement toward nutrients.

  • Biofilm Formation Studies: Supports controlled biofilm assays for Pseudomonas aeruginosa and Escherichia coli on a minimal carbon source.

  • Induction Optimization for Expression Systems: Facilitates tuning of IPTG or arabinose induction in a nutrient-defined background.
