M9 Minimal Salts Agar Plates with Uracil-250 is a defined minimal agar medium for cultivation and analysis of bacteria such as E. coli under controlled nutrient conditions. Glucose serves as the primary carbon and energy source, thiamine supplies vitamin B1, and magnesium and calcium provide essential divalent cations. The addition of uracil (250 µg/mL) supports the growth of uracil-auxotrophic strains on a solid minimal matrix.

  • 1X M9 Salts
  • 1.0% Cerelose Anhydrous Dextrose (Glucose)
  • 1.0mM Thiamine
  • 0.2mM Magnesium Sulfate
  • 0.1mM Calcium Chloride
  • 1.5% Agar
  • 250µg/mL Uracil
  • Growth of Uracil Auxotrophs: Supports maintenance and propagation of pyrE/pyrF mutants of Escherichia coli and related enterics with 250µg/mL uracil.

  • Defined Minimal Culture: Enables controlled growth of E. coli on 1X M9 salts with 1.0% glucose for physiology and metabolism studies.

  • Auxotrophy-Based Selection/Complementation: Facilitates selection and verification of pyrimidine biosynthesis mutants and complemented strains on minimal medium.

  • Bacteriophage Plaque Assays: Suitable for phage work (e.g., P1, T4, λ) on E. coli with 0.2mM MgSO₄ and 0.1mM CaCl₂ enhancing adsorption.

  • Carbon Utilization and CCR Assays: Tests glucose utilization and catabolite repression under defined 1.0% glucose conditions.

  • Recombineering and Genome Editing Workflows: Enables selection schemes involving pyrF/pyrE counterselection complemented by uracil.
