M9 Minimal Salts Agar Plates with Kanamycin-50 are a defined minimal agar medium used for growth and selection of kanamycin-resistant E. coli strains under carbon-limited conditions.

Glucose serves as the sole carbon source, thiamine provides an essential vitamin supplement, and magnesium and calcium supply required divalent cations. Kanamycin sulfate at 50 µg/mL enables selection of plasmid-bearing transformants.

  • 1X M9 Salts
  • 1.5% Agar
  • 1.0% Cerelose Anhydrous Dextrose (Glucose)
  • 1.0mM Thiamine
  • 0.2mM Magnesium Sulfate
  • 0.1mM Calcium Chloride
  • 50µg/mL Kanamycin Sulfate
  • Selection of Kanamycin-Resistant Transformants: Isolation and maintenance of E. coli clones carrying kanamycin-resistant plasmids at 50 µg/mL.

  • Recombinant Colony Screening: Post-transformation colony picking and verification under kanamycin selection.

  • Conjugation/Transduction Selection: Recovery of kanamycin-marked alleles or transposon insertions in E. coli.

  • CRISPR/Cas Editing Selection: Enrichment of edited E. coli clones using kanamycin resistance markers.

  • Defined Minimal Growth Studies: Metabolism, auxotrophy, and fitness assays on M9 with glucose as the sole carbon source.

  • Expression Studies in Minimal Media: Controlled background for gene expression from kanamycin-selectable constructs.

  • Plasmid Stability Assays: Monitoring retention of kanamycin-selectable plasmids under continuous selection.

  • Colony Purification and Storage: Streaking and short-term storage of kanamycin-resistant E. coli strains on 1.5% agar.

  • Antibiotic Marker Verification: Confirmation of kanamycin resistance phenotypes in laboratory strains.
