M9 Minimal Salts Agar Plates are a defined minimal agar medium for cultivation, maintenance, and genetic screening of enteric bacteria such as E. coli under controlled nutrient conditions.

The formulation provides a single glucose carbon source, thiamine supplementation, essential Mg2+ and Ca2+ ions, and an agar solid matrix, supporting growth on defined nutrients and differentiation based on metabolic capabilities.

  • 1X M9 Salts
  • 1.0% Cerelose Anhydrous Dextrose (Glucose)
  • 1mM Thiamine
  • 0.2mM Magnesium Sulfate
  • 0.1mM Calcium Chloride
  • 1.5% Agar
  • Selection of Auxotrophs/Prototrophs: Defined 1X M9 with 1.0% glucose and thiamine enables genetic selection for biosynthetic capabilities in E. coli and related bacteria.

  • Metabolic and Carbon Utilization Studies: Glucose-based minimal agar supports phenotyping of central carbon metabolism and substrate utilization in E. coli.

  • Antibiotic Selection on Defined Medium: Maintains controlled ionic conditions (Mg2+, Ca2+) for plasmid maintenance and selection with antibiotics in E. coli.

  • Replica Plating for Nutritional Requirements: Used to map auxotrophies and screen revertants or suppressors on minimal plates.

  • Bacteriophage Assays (Base Plate): Serves as the bottom agar for soft-agar overlays in phage plaque assays with E. coli and coliphages.

  • CFU Enumeration and Colony Isolation: Supports counting and isolating colonies of organisms adapted to minimal media, including E. coli.

  • Conjugation and Transduction Selections: Facilitates selection of recombinants using auxotrophic markers during Hfr conjugation or P1 transduction in E. coli.

  • Defined-Medium Culturing for Systems Biology: Provides reproducible growth conditions for transcriptomics/proteomics of E. coli under minimal nutrient regimes.

  • Growth of Non-Fastidious Model Bacteria: Supports cultivation of minimal-medium-competent strains such as E. coli and some Pseudomonas spp.
