MacConkey Agar Plates are a selective and differential medium for the isolation and differentiation of Gram-negative enteric bacteria based on lactose fermentation. Bile salts and crystal violet suppress Gram-positive organisms, while lactose and neutral red enable visual distinction of lactose-fermenting colonies (pink to red, often with bile precipitate) from non-lactose fermenters (colorless to translucent).

Tryptone and peptone supply amino acids and peptides, sodium chloride maintains osmotic balance, and agar provides a solid growth surface. Typical applications include primary isolation and differentiation of organisms such as Escherichia coli, Enterobacter, and Klebsiella (lactose fermenters) from non-fermenters such as Salmonella and Shigella.

  • 1.7% Tryptone
  • 0.3% Peptone
  • 1.0% Lactose
  • 0.15% Bile Salts #3
  • 0.5% Sodium Chloride
  • 1.35% Agar
  • 30µg/mL Neutral Red
  • 1µg/mL Crystal Violet
  • Isolation of Enteric Gram-Negatives: Selective recovery of Gram-negative rods from clinical specimens (urine, stool, wound) while suppressing Gram-positives.

  • Differentiation by Lactose Fermentation: Distinguishes lactose fermenters (pink/red colonies) such as Escherichia coli, Klebsiella, and Enterobacter from non-fermenters (colorless) like Salmonella, Shigella, and Pseudomonas aeruginosa.

  • Coliform Detection: Screening and enumeration of coliforms in water, food, and dairy testing based on lactose fermentation and bile tolerance.

  • Clinical Microbiology Workflows: Primary plating and subculture to guide identification and downstream biochemical or MALDI-TOF testing of Enterobacterales.

  • Public Health Surveillance: Recovery of enteric pathogens (e.g., non-lactose-fermenting Salmonella and Shigella) from stool and enrichment broths.

  • Environmental Monitoring: Detection of Gram-negative contaminants and coliforms in pharmaceutical and cleanroom surveillance programs.

  • Mixed Culture Isolation: Selective plating from polymicrobial specimens to reduce overgrowth by Gram-positives and fastidious flora.

  • Quality Control and Validation: Routine QC with reference strains to verify selective and differential performance for lactose fermentation and bile tolerance.
