LB Broth is a nutrient-rich liquid medium used for the cultivation and maintenance of E. coli and other nonfastidious bacteria in molecular biology and microbiology workflows.

The peptone and yeast extract provide amino acids, peptides, vitamins, and trace elements, and sodium chloride maintains ionic strength and osmotic balance. Common applications include routine culture propagation, strain maintenance, and preparation of cells for downstream analyses.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • Routine Bacterial Cultivation: General growth and propagation of non-fastidious organisms such as Escherichia coli using a classic 1.0% Tryptone, 0.5% Yeast Extract, 1.0% NaCl formulation.

  • Starter and Overnight Cultures: Inoculum preparation for downstream cloning, expression, and screening workflows.

  • Plasmid Amplification: Growth of E. coli for mini-, midi-, and maxi-prep DNA isolation.

  • Post-Transformation Recovery: Short-term outgrowth after chemical transformation or electroporation prior to selection.

  • Antibiotic Selection in Liquid Culture: Selection and maintenance of recombinant strains when supplemented with antibiotics (e.g., ampicillin, kanamycin).

  • Pre-Cultures for Protein Expression: Seed cultures used to inoculate richer expression media or main fermentation runs.

  • LB Agar Plate Preparation: Base medium for solid media when combined with agar for colony isolation and selection.

  • Bacteriophage Workflows: Growth of host bacteria and overlays for phage propagation and titering.

  • Strain Maintenance: Routine culturing prior to preparing glycerol stocks or cryopreservation.
