LB Broth with Carbenicillin-100 is a lysogeny broth medium for cultivation and selection of carbenicillin-resistant E. coli strains. The inclusion of 100 µg/mL carbenicillin disodium salt provides selective pressure to maintain beta-lactam–resistance plasmids in plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 100µg/mL Carbenicillin Disodium Salt
  • Plasmid Selection in E. coli: Liquid culture growth of transformants carrying beta-lactamase (ampR) plasmids using 100 µg/mL carbenicillin for selection.

  • Transformation Recovery and Expansion: Outgrowth and overnight cultures of recombinant E. coli following heat-shock or electroporation under beta-lactam selection.

  • Plasmid DNA Preparation: Culturing E. coli for miniprep, midiprep, or maxiprep workflows to isolate plasmid DNA.

  • Protein Expression Starter Cultures: Inoculation and growth of pre-induction seed cultures of recombinant E. coli harboring carbenicillin-selectable expression vectors.

  • Glycerol Stock Preparation: Generating long-term freezer stocks of validated E. coli clones under carbenicillin selection.

  • Strain and Plasmid Maintenance: Routine propagation and passage of plasmid-bearing E. coli to preserve constructs and prevent plasmid loss.

  • Library Amplification: Expansion of plasmid libraries in E. coli where beta-lactam selection is required to maintain library diversity.

  • Post-Colony Screening Cultures: Growing verified colonies in liquid LB with carbenicillin for downstream assays, sequencing, or functional testing.
