LB Broth with Carbenicillin-50 is a nutrient medium for cultivating and selecting carbenicillin-resistant E. coli strains in liquid culture. Carbenicillin disodium salt at 50 µg/mL provides beta-lactam selection pressure to maintain plasmids carrying beta-lactamase markers during growth and propagation.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 50µg/mL Carbenicillin Disodium Salt
  • Antibiotic Selection of Plasmid-Bearing Escherichia coli: Maintains AmpR/bla plasmids using 50 µg/mL carbenicillin in a 1.0% Tryptone, 0.5% Yeast Extract, 1.0% NaCl LB base.

  • Transformation Recovery and Outgrowth: Supports post-transformation growth of Escherichia coli prior to plating under beta-lactam selection.

  • Plasmid DNA Amplification: Culturing AmpR vectors in Escherichia coli for mini/midi/maxi-prep workflows.

  • Recombinant Protein Expression: Shake-flask growth of AmpR expression constructs in Escherichia coli with stable carbenicillin selection.

  • Starter Cultures and Scale-Up: Inoculum preparation and culture expansion for downstream fermentation with beta-lactam selection.

  • Glycerol Stock Preparation: Long-term archiving of transformed Escherichia coli under 50 µg/mL carbenicillin pressure.

  • Blue-White Screening (with Supplements): Used with X-gal/IPTG for α-complementation screening of AmpR cloning vectors in Escherichia coli.

  • Maintenance of Phagemids and Shuttle Vectors: Propagation of AmpR phagemids/shuttle plasmids in Escherichia coli.

  • Library Construction and Enrichment: Growth and selection of plasmid libraries (e.g., mutagenesis or variant libraries) in Escherichia coli with beta-lactam resistance markers.

  • Quality Control of AmpR Constructs: Routine culturing for plasmid stability testing and insert verification in Escherichia coli.

  • pH: 7.0 ± 0.10
