LB Animal-Free Agar is a nutritionally balanced agar medium formulated with plant-derived peptone, yeast extract, sodium chloride, and agar. It is intended for routine cultivation, maintenance, and plating of non-fastidious bacteria, including laboratory strains of E. coli. Sodium chloride maintains osmotic balance, and agar provides a solid matrix for colony isolation and enumeration. Antibiotics or other selective agents may be added as required for specific applications.

  • 1.69% Animal-Free Soy Peptone
  • 0.50% Yeast Extract
  • 1.00% Sodium Chloride
  • 1.50% Agar
  • Bacterial Colony Isolation and Enumeration: Plating and CFU counting of non-fastidious bacteria such as Escherichia coli, Bacillus subtilis, and Pseudomonas putida.

  • Cloning and Plasmid Selection: Growth of transformants on selective plates (e.g., ampicillin, kanamycin) for plasmid maintenance in Escherichia coli.

  • Transformant Recovery and Screening: Post-transformation plating and colony screening, including blue-white screening when supplemented with X-gal/IPTG.

  • Strain Maintenance: Short-term storage and routine streaking of laboratory strains of Escherichia coli and other non-fastidious bacteria.

  • Recombinant Protein Workflow Support: Colony screening prior to liquid culture for expression studies in Escherichia coli.

  • Phage Assays (with Soft Agar Overlay): Preparation of bacterial lawns for plaque assays when used alongside a compatible soft agar overlay.

  • Replica Plating and Phenotypic Selection: Screening for auxotrophies or selectable markers on supplemented LB Animal-Free plates.

  • Microbial QC and Environmental Monitoring: Bioburden checks and indicator organism recovery in animal-free workflows.

  • Teaching and Training: Educational microbiology exercises requiring an animal-free medium.

  • Animal-Free Process Development: Culture of production or chassis strains under animal-component–free conditions for regulatory or ethical compliance.
