LB Broth is a nutrient medium for cultivation and maintenance of E. coli and other bacteria in routine culture workflows. It provides peptides and amino acids from tryptone, vitamins and growth factors from yeast extract, and sodium chloride to maintain osmotic balance.

Applications include propagation of plasmid-bearing strains, starter and overnight cultures, and general microbiological procedures. The medium can be supplemented with antibiotics or carbohydrates as required and is typically sterilized by autoclaving.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • Routine Bacterial Cultivation: General growth and propagation of Escherichia coli using the standard LB Miller formulation (1.0% Tryptone, 0.5% Yeast Extract, 1.0% NaCl).

  • Plasmid Cloning and Maintenance: Culturing recombinant Escherichia coli for plasmid construction, maintenance, and selection with antibiotics.

  • Transformation Recovery/Outgrowth: Post-heat shock or electroporation recovery of Escherichia coli prior to plating or selection.

  • Starter Cultures for DNA Prep: Overnight cultures of Escherichia coli for miniprep, midiprep, and maxiprep workflows.

  • Protein Expression Cultures: Growth of recombinant Escherichia coli for IPTG- or autoinduction-based protein expression.
