LB Agar with Carbenicillin-100, Kanamycin-50, Tetracycline-10, Bluo-gal-100, and IPTG-40 is a Luria–Bertani agar medium that supports the growth and selection of carbenicillin-, kanamycin-, and tetracycline-resistant E. coli transformants. The inclusion of IPTG and Bluo-gal enables blue/white screening through induction of lacZ α-complementation and chromogenic detection of β-galactosidase activity.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin
  • 50µg/mL Kanamycin
  • 10µg/mL Tetracycline
  • 100µg/mL Bluo-Gal
  • 40µg/mL IPTG
  • Blue/White Colony Screening: Alpha-complementation of lacZ using IPTG (40 µg/mL) and Bluo-Gal (100 µg/mL) for rapid identification of recombinant clones in E. coli.

  • Tri-Antibiotic Selection: Concurrent selection for plasmids bearing carbenicillin (100 µg/mL), kanamycin (50 µg/mL), and tetracycline (10 µg/mL) resistance markers in E. coli.

  • Multi-Plasmid Maintenance: Stable co-maintenance of compatible vectors with distinct resistance cassettes in cloning and expression strains of E. coli.

  • Library Construction and Screening: High-throughput colony picking and screening of ligation, Gibson, or Golden Gate libraries under triple selection in E. coli.
