LB Agar Plates with Carbenicillin-100 is a solid agar medium for cultivation and selection of carbenicillin-resistant E. coli strains. The LB agar base supplies nutrients and ionic balance, and 100 µg/mL carbenicillin enables selection of plasmid-bearing transformants.

Suitable for routine cloning, colony screening, and maintenance of strains carrying beta-lactam resistance plasmids in E. coli.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin
  • Antibiotic Selection of Transformants: Selects for bla-encoded carbenicillin resistance in Escherichia coli and other Gram-negative hosts after transformation.

  • Plasmid Maintenance: Maintains plasmids carrying beta-lactamase by continuous selection during strain propagation and streaking for isolation.

  • High-Density Colony Screening: Uses 245 mm gridded format for arraying, colony picking, and library screening on large-format plates.

  • CFU Enumeration: Enables accurate colony counting across serial dilutions using the gridded surface.

  • Replica Plating and Genetic Screens: Facilitates replica plating of arrays to assess phenotypes while retaining carbenicillin selection.

  • Cloning Workflows: Supports selection of recombinant clones; carbenicillin reduces satellite colonies compared to ampicillin.

  • Blue-White Screening: Suitable for alpha-complementation assays (with X-gal/IPTG) to identify inserts under carbenicillin selection.

  • Conjugation/Mating Selections: Selects transconjugants carrying bla-marked plasmids following bacterial mating.

  • Strain and Expression Line Maintenance: Preserves expression constructs prior to starter culture setup for protein production in Escherichia coli.

  • Plasmid Stability Testing: Assesses segregational stability of bla-marked plasmids under antibiotic pressure.
