LB Agar Plates with Carbenicillin-50 are a lysogeny broth–based agar medium that supports the growth and selection of carbenicillin-resistant E. coli strains. The inclusion of 50 µg/mL carbenicillin disodium salt enables selection of plasmid-bearing transformants on solid medium.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Carbenicillin Disodium Salt
  • Plasmid Selection in Escherichia coli: Selects and maintains bla/ampR plasmid-bearing transformants using 50 µg/mL carbenicillin; minimizes satellite colonies compared to ampicillin.

  • Colony Screening and Picking: Plates cloning reactions and transformations to isolate single colonies for downstream analysis.

  • Blue-White Screening: When supplemented with X-gal/IPTG, supports lacZ α-complementation for insert screening in E. coli.

  • Library Construction and Screening: Plating and selection of combinatorial, mutagenesis, or genomic libraries carrying bla-marked vectors.

  • CFU Enumeration and Transformation Efficiency: Quantifies colony-forming units and transformation efficiency of competent E. coli.

  • Strain Maintenance and Archiving: Streaking and short-term storage of carbenicillin-resistant E. coli strains on solid LB.

  • Conjugation/Mating Selection: Selects transconjugants carrying beta-lactamase plasmids after bacterial mating assays.

  • CRISPR/Genome Editing Workflows: Screens edited clones maintained on bla-selectable cloning vectors in E. coli.

  • Phagemid/Cloning Vector Recovery: Plating of ligations or phagemid rescue workflows using bla/ampR vectors.

  • Starter Culture Initiation for Expression: Picking verified colonies into liquid media for small-scale expression prescreens.
