LB Agar Plates with Tetracycline-10 is a lysogeny broth agar medium that supports the growth and selection of tetracycline-resistant E. coli strains. The inclusion of 10 µg/mL tetracycline hydrochloride enables selection and maintenance of plasmid-bearing transformants carrying tetracycline resistance markers.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 10µg/mL Tetracycline Hydrochloride
  • Selective Isolation of Tetracycline-Resistant Colonies: Recovery of transformants carrying tet-marked plasmids on LB agar (1.0% Tryptone, 0.5% Yeast Extract, 1.0% Sodium Chloride, 1.5% Agar) with 10µg/mL Tetracycline Hydrochloride.

  • Plasmid Maintenance and Propagation: Stable maintenance of tetracycline-resistance plasmids in E. coli strains.

  • Bacterial Cloning and Subcloning: Screening recombinant constructs that use tetracycline resistance as the selectable marker.

  • Conjugation and Transposon Selection: Selection of transconjugants or transposon insertions conferring tetracycline resistance.

  • Strain Verification: Confirming genotype/phenotype of tetracycline-resistant strains compared to non-resistant controls.

  • Colony Isolation and Purification: Streaking for single colonies from mixed cultures under tetracycline selection.

  • CFU Enumeration Under Selection: Quantifying transformation or conjugation efficiency by plating dilutions on tetracycline plates.

  • Maintenance of Recombinant Expression Hosts: Culturing tet-marked expression vectors in E. coli for downstream protein or DNA preparation.

  • Library and Mutant Collection Handling: Propagation of genomic, cDNA, or transposon libraries using tetracycline selection.

  • Educational and QC Applications: Teaching antibiotic selection principles and routine QC of tetracycline marker function in lab strains.
