LB Agar Plates with Kanamycin-50 are solid LB medium designed for cultivation and selection of kanamycin-resistant E. coli strains. The agar matrix supports discrete colony formation for routine screening and strain maintenance.

Kanamycin sulfate at 50 µg/mL provides selection pressure for plasmid-bearing transformants, enabling maintenance of kanamycin resistance markers during cloning and propagation workflows.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Kanamycin Sulfate
  • Antibiotic Selection of Transformants: Selects kanamycin-resistant colonies (e.g., plasmid-bearing E. coli) after transformation.

  • Colony Isolation and Purification: Streaking for single colonies under kanamycin to obtain clonal, plasmid-maintained isolates.

  • Plasmid Maintenance and Propagation: Routine subculture of strains carrying kanamycin-selectable plasmids to preserve selection.

  • Colony PCR/Screening: Picking kanamycin-resistant colonies for PCR, restriction analysis, or sequencing-based clone verification.

  • Transformation Efficiency and CFU Enumeration: Plating to quantify viable kanamycin-resistant transformants per μg DNA.

  • Genetic Engineering Selection: Selection of recombineering, CRISPR/Cas9 edits, or insertion events marked by kanamycin resistance in E. coli.

  • Conjugation/Transduction Assays: Selecting kanamycin-marked transconjugants or transductants in bacterial genetics workflows.

  • Library Plating and Screening: Amplification and screening of plasmid or mutagenesis libraries carrying kanamycin resistance.

  • Replica Plating and Stability Testing: Assessing plasmid retention and fitness under kanamycin selection pressure.

  • Strain Stocking and Short-Term Storage: Maintaining selective plate stocks of engineered E. coli strains.
