LB Agar Plates with Carbenicillin-100 are solid media for cultivation and selection of carbenicillin-resistant E. coli strains. The inclusion of 100 µg/mL carbenicillin enables selection and maintenance of beta-lactam–selectable plasmids in transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/ml Carbenicillin
  • Antibiotic Selection of Plasmid-Bearing Clones: Selects E. coli carrying bla/ampR plasmids using 100 µg/ml carbenicillin for robust colony growth.

  • Bacterial Transformation and Cloning: Plates heat-shock or electroporated E. coli to recover and select transformants.

  • Colony Screening (Colony PCR/Sanger Prep): Grows discrete colonies for downstream PCR, minipreps, and sequencing.

  • Blue-White Screening: When supplemented with X-Gal/IPTG, enables lacZ α-complementation screening on carbenicillin selection.

  • Strain Maintenance and Streaking: Maintains selection pressure for plasmid retention during routine passaging of E. coli.

  • Plasmid Library Propagation: Supports high-complexity library plating and colony picking on 245 mm low-profile plates.

  • Recombinant Expression Prescreening: Picks verified carbenicillin-resistant colonies for small-scale expression tests.

  • Glycerol Stock Preparation: Isolates single resistant colonies for stable long-term strain archiving.

  • Conjugation/Mating with Selection: Recovers transconjugants in E. coli using carbenicillin-selectable plasmids.

  • Reduction of Satellite Colonies: Uses carbenicillin to minimize satellites compared to ampicillin for cleaner selection.
