LB Agar Plates with Carbenicillin-100 are a nutrient agar medium that supports the growth and selection of carbenicillin-resistant E. coli strains. The solid LB matrix facilitates colony isolation and routine cultivation, and 100 µg/mL carbenicillin disodium salt enables selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin Disodium Salt
  • Plasmid Selection and Maintenance: Selects and maintains bla (beta-lactam) resistance plasmids in Escherichia coli using 100 µg/mL carbenicillin.

  • Bacterial Transformation Screening: Recovers and isolates transformants on selective LB agar after heat shock or electroporation of Escherichia coli.

  • Colony Isolation and Purification: Streaks single colonies to ensure clonal, antibiotic-resistant cultures of Escherichia coli.

  • Colony PCR and Insert Screening: Picks antibiotic-selected colonies for PCR verification of inserts or edits.

  • Recombinant Protein Workflow Support: Maintains expression strains harboring bla-marked vectors prior to induction in liquid culture.

  • Library Construction and Screening: Selects cDNA/genomic libraries and variant libraries maintained on beta-lactam markers.

  • Blue-White Screening (with supplements): Supports alpha-complementation screening on carbenicillin plates when supplemented with X-Gal and IPTG.

  • CFU Enumeration Under Selection: Quantifies viable antibiotic-resistant cells after cloning, mutagenesis, or selection steps.

  • Plasmid Transfer Selection: Selects transconjugants or transductants carrying bla-marked plasmids in Escherichia coli.

  • Strain Archiving and Stability Checks: Verifies plasmid retention and phenotype stability by restreaking from glycerol stocks under carbenicillin selection.
