LB Agar Plates 1% Glucose and Kanamycin-50 is a nutrient-rich agar medium that supports the growth and selection of kanamycin-resistant E. coli strains. The addition of 1% glucose enhances metabolic activity and biomass yield, and 50 µg/mL kanamycin sulfate enables effective selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 1.0% Glucose
  • 50µg/mL Kanamycin Sulfate
  • Antibiotic Selection of Transformants: Isolation of kanamycin-resistant colonies after plasmid transformation in Escherichia coli/E. coli.

  • Plasmid Maintenance: Routine propagation of kanamycin-marked plasmids in E. coli under 50 µg/mL kanamycin selection.

  • Colony Isolation and Purification: Streaking and single-colony purification on LB 1% glucose with kanamycin.

  • CFU Enumeration: Quantifying viable kanamycin-resistant cells via spread plating.

  • High-Density Library Plating: 150 mm plates for screening mutant/genomic libraries under kanamycin selection.

  • Replica Plating: Verification of kanamycin resistance and genotype confirmation across selective media.

  • Conjugation/Transposon Selections: Selecting transconjugants or insertions carrying kanamycin resistance in E. coli.

  • CRISPR/Cas Workflows: Selection of editing plasmids with kanamycin markers in E. coli.

  • Reduced Basal Expression: Using 1% glucose to suppress leaky expression from lac/T7–lac systems prior to induction.

  • Competent Cell QC: Transformation efficiency testing with kanamycin-resistance control plasmids.

  • Short-Term Archiving: Preparing working stocks and master plates of kanamycin-resistant strains.
