LB Agar Plates with 1% Glucose, Carbenicillin-100 is a nutrient agar medium that supports the growth and selection of carbenicillin-resistant E. coli strains. The addition of 1% glucose enhances metabolic activity and biomass yield, and 100 µg/mL carbenicillin enables selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast extract
  • 1.0% Sodium chloride
  • 1.5% Agar
  • 1.0% Cerelose Anhydrous Dextrose (Glucose)
  • 100µg/mL Carbenicillin
  • Antibiotic Selection of Transformants: Isolation of carbenicillin-resistant Escherichia coli colonies carrying bla-based plasmids at 100 µg/mL carbenicillin.

  • Plasmid Maintenance and Clone Stabilization: Routine passaging of bla-selectable plasmids in E. coli, with carbenicillin providing stable beta-lactam selection.

  • Colony Screening and Picking: Growth of single colonies for downstream minipreps, PCR colony screening, and sequence verification.

  • Transformation Recovery Plating: Post-transformation plating to quantify CFUs and calculate transformation efficiency under beta-lactam selection.

  • Streaking for Isolation: Single-colony isolation from mixed cultures or glycerol stocks of E. coli to ensure clonal purity.
