LB Agarose Plates with Kanamycin-50 are a solid LB medium for cultivation and selection of kanamycin-resistant E. coli strains. The inclusion of 50 µg/mL kanamycin sulfate enables selection of plasmid-bearing transformants on a firm agarose matrix suitable for colony isolation and routine propagation.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agarose
  • 50µg/mL Kanamycin Sulfate

  • Selection of Kanamycin-Resistant Transformants: Isolates colonies of Escherichia coli/E. coli carrying kanamycin-resistant plasmids.

  • Plasmid Maintenance and Propagation: Sustains cosmids/BACs/standard plasmids with kan markers in E. coli.

  • Colony Screening: Enables colony PCR or replica plating of kan-selected clones.

  • Transformation Efficiency Assays: Quantifies CFUs under 50 µg/mL kanamycin selection.

  • Transposon/Insertion Mutant Selection: Selects kan-marked mutants in E. coli, Bacillus subtilis, and Agrobacterium tumefaciens (where kanamycin is effective).

  • Conjugation/Mating Selections: Recovers donors/recipients with kan markers after biparental or triparental matings in E. coli workflows.

  • Library Construction and Screening: Plates genomic/cDNA libraries with kan-selectable vectors.

  • Strain Archiving and Maintenance: Streaks and short-term stores kan-resistant strains on solid LB agarose.

  • Recovery from Frozen Stocks: Revives glycerol stocks onto selective plates to prevent plasmid loss.

  • Quality Control of Selectable Markers: Verifies kan-resistance cassette functionality in engineered strains.
