LB Lennox Agar Plates with Blasticidin-100, pH 8 is a solid nutrient agar medium for cultivation and selection of blasticidin-resistant E. coli and other bacteria. The formulation supports routine growth on plates while providing conditions appropriate for screening transformants.

The medium is adjusted to pH 8 and contains 100 µg/mL Blasticidin S for selection of cells harboring a blasticidin-resistance marker. Supplied as ready-to-use agar plates for microbial culture and plasmid-based selection workflows.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Blasticidin S
  • Selection of Blasticidin-Resistant Transformants: Plating transformed E. coli to select colonies carrying plasmids with bsd/bsr resistance at 100 µg/mL blasticidin.

  • Plasmid Maintenance and Propagation: Routine streaking and passaging of recombinant E. coli strains to maintain blasticidin-selectable plasmids.

  • Colony Screening and Verification: Picking colonies for colony PCR, restriction analysis, or sequencing from blasticidin-selective plates.

  • CRISPR/Cas9 and Genome Editing Workflows: Selecting edited bacterial clones carrying blasticidin-marked editing or helper plasmids.

  • Protein Expression Clone Selection: Isolating expression strains with blasticidin-resistant expression vectors prior to small-scale or large-scale induction.

  • Library Construction and Clonal Isolation: Selecting and isolating clones from plasmid or mutagenesis libraries using blasticidin resistance in E. coli.

  • Antibiotic Susceptibility/Resistance Confirmation: Verifying functional blasticidin resistance markers in engineered bacterial strains.

  • Cross-Contamination Control in Mixed Cultures: Suppressing growth of non-resistant background microbes while recovering target blasticidin-resistant colonies.

  • Alternative Host Selection (as applicable): Selecting blasticidin-resistant transformants in other amenable bacteria (e.g., Bacillus subtilis) when a compatible bsd/bsr marker is present.
