LB Lennox Agar with Carbenicillin-100 is an agar medium used for cultivation and selection of carbenicillin-resistant E. coli strains. The inclusion of 100 µg/mL carbenicillin disodium salt enables selection of plasmid-bearing transformants on solid medium.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 0.5% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin Disodium Salt
  • Selective Plating of Transformants: Recovery and selection of plasmid-bearing E. coli on 100 µg/mL carbenicillin after transformation.

  • Plasmid Maintenance and Propagation: Routine culture of beta-lactam–resistant E. coli strains to maintain plasmids with bla/ampR.

  • Colony Isolation and CFU Enumeration: Streaking for single colonies and counting viable cells on LB Lennox agar.

  • Cloning and Screening Workflows: Colony picking and insert verification; compatible with blue/white screening when supplemented with X-gal/IPTG.

  • Expression Construct Screening: Selecting and prescreening recombinant expression clones in E. coli prior to induction.

  • Glycerol Stock Revival: Reviving stored strains onto selective plates to ensure plasmid retention.

  • Conjugation/Mating Selections: Selecting transconjugants carrying beta-lactam–resistant plasmids on Gram-negative hosts.

  • Strain Verification and Stability Testing: Assessing plasmid stability and segregation under beta-lactam selection pressure.

  • General Gram-Negative Selection: Selection of susceptible non-enteric Gram-negatives (e.g., Pseudomonas aeruginosa) carrying carbenicillin-resistance markers.
