LB Agar Plates with Carbenicillin-200 is an LB agar medium that supports the growth and selection of carbenicillin-resistant E. coli strains. The inclusion of 200 µg/mL carbenicillin disodium salt provides selection pressure for maintenance of beta-lactamase–encoding plasmids and recovery of transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 200µg/mL Carbenicillin Disodium Salt
  • Selective Plating of bla-Encoded Plasmids: Enables antibiotic selection of beta-lactamase plasmids in E. coli using 200 µg/mL carbenicillin.

  • Bacterial Transformation and Colony Isolation: Used for post-transformation recovery and isolation of individual recombinant colonies in E. coli.

  • Plasmid Maintenance and Strain Purification: Supports streaking and passaging to maintain plasmids and obtain clonal isolates under beta-lactam selection.

  • Colony Forming Unit (CFU) Enumeration: Facilitates quantification of viable, plasmid-bearing cells under selective pressure.

  • Colony PCR Screening: Provides selective growth of transformants for downstream colony PCR verification of inserts.

  • Library Construction and Screening: Used to plate and screen recombinant DNA libraries while maintaining selection for bla-marked vectors.

  • Starter Culture Prep for Protein Expression: Enables picking verified colonies for selective overnight cultures prior to expression in E. coli.

  • Reduced Satellite Colonies vs. Ampicillin Plates: Carbenicillin helps minimize satellite colonies during selection relative to ampicillin.

  • Selection in Alternative Hosts: Applicable to other susceptible Gram-negative bacteria (e.g., Agrobacterium tumefaciens, Pseudomonas) carrying carbenicillin-resistant plasmids.

  • Conjugation/Transconjugant Selection: Used to select for transconjugants that acquire bla-bearing plasmids after mating experiments.
