LB Agar Plates with Kanamycin-50 are a nutrient agar medium that supports the growth and selection of kanamycin-resistant E. coli strains. Inclusion of 50 µg/mL kanamycin sulfate enables selection of plasmid-bearing transformants on a solid surface.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Kanamycin Sulfate
  • Antibiotic Selection of Transformants: Selects kanamycin-resistant colonies after cloning and transformation of Escherichia coli (50 µg/mL Kan) on LB (1.0% Tryptone, 0.5% Yeast Extract, 1.0% NaCl, 1.5% Agar).

  • Plasmid Maintenance and Propagation: Maintains vectors carrying kanR in laboratory strains of E. coli and other compatible bacteria under selective pressure.

  • Colony Isolation and Purification: Streaks for single colonies to purify kanamycin-resistant strains prior to downstream experiments.

  • Colony PCR and Insert Screening: Provides colonies for PCR verification, restriction analysis, or Sanger sequencing of kan-resistant clones.

  • Transformation Efficiency and CFU Enumeration: Plates serial dilutions to quantify transformants (CFU) under kanamycin selection.

  • Protein Expression Workflows: Plates expression strains harboring kan-resistant plasmids prior to induction and small-scale culture setup.

  • Genome Editing and Recombineering Selection: Selects recombinants carrying kanamycin cassettes used in gene knockouts or insertions.

  • Conjugation/Transduction/Transformation Selection: Recovers recipients acquiring kan-marked elements after mating or phage-mediated transfer.

  • Library Construction and Screening: Screens mutant or genomic libraries where constructs confer kanamycin resistance.

  • Strain Maintenance and Stock Preparation: Verifies viability and resistance of strains (e.g., Escherichia coli, Salmonella enterica, Bacillus subtilis) before creating glycerol stocks.
