LB Agar Plates with Chloramphenicol-12.5 is a nutrient agar medium that supports the growth and selection of chloramphenicol-resistant E. coli strains. Inclusion of 12.5 µg/mL chloramphenicol enables selection of transformants and maintenance of plasmids bearing chloramphenicol resistance markers.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 12.5µg/mL Chloramphenicol
  • Selection of Chloramphenicol-Resistant Transformants: Isolates colonies carrying chloramphenicol-selectable plasmids or genomic inserts in Escherichia coli.

  • Plasmid and BAC/PAC/Cosmid Maintenance: Propagates chloramphenicol-selectable vectors and libraries in Escherichia coli at 12.5 µg/mL.

  • Colony Screening and Picking: Enables selection-based cloning workflows (e.g., miniprep prep, Sanger prep) for chloramphenicol-marked constructs.

  • CFU Enumeration Under Selection: Quantifies viable chloramphenicol-resistant cells after transformation, conjugation, or transduction.

  • Replica Plating for Knockout Verification: Screens gene disruptions using cat (chloramphenicol acetyltransferase) cassettes in Escherichia coli.

  • Transposon Mutagenesis Selection: Recovers chloramphenicol-marked transposon insertions in Escherichia coli and Salmonella enterica.

  • CRISPR Editing Workflows: Selects edits that retain or integrate chloramphenicol resistance markers.

  • Conjugation and Mating Selections: Selects recipients or exconjugants acquiring chloramphenicol-marked plasmids.

  • Strain Maintenance Under Antibiotic Pressure: Maintains chloramphenicol-resistant lab strains of Escherichia coli and other bacteria (e.g., Bacillus subtilis).

  • High-Density Colony Arrays on 150 mm Plates: Supports large-scale screening and library plating with chloramphenicol selection.
