LB Agar Plates with Carbenicillin-100 are nutrient-rich agar media that support the growth and selection of carbenicillin-resistant E. coli strains. Luria-Bertani nutrients provide general cultivation conditions, and 100 µg/mL carbenicillin disodium salt enables selection of plasmid-bearing transformants carrying beta-lactam resistance markers.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin Disodium Salt
  • Antibiotic Selection of Transformants: Isolates colonies carrying bla/ampR plasmids on 100 µg/mL carbenicillin LB agar, commonly in Escherichia coli.

  • Clonal Isolation (Streaking for Singles): Generates single, genetically uniform colonies from mixed or transformed cultures.

  • Colony PCR Screening: Provides colonies as templates to verify inserts or constructs directly from carbenicillin-selected plates.

  • CFU Enumeration and Viability: Quantifies transformation efficiency and cell viability under beta-lactam selection.

  • Plasmid Maintenance and Stability Testing: Maintains selection pressure to assess plasmid retention across passages.

  • Strain Recovery from Stocks: Revives and amplifies stored carbenicillin-resistant strains (e.g., Escherichia coli) for downstream culture.

  • Pre-culture Plate Setup for Cloning/Expression: Prepares verified colonies for inoculation into liquid media with beta-lactam selection.

  • Conjugation/Mating Selections: Selects transconjugants carrying beta-lactam–resistant plasmids after biparental or triparental matings.
