LB Agar Plates with Ampicillin-100 are solid nutrient medium designed for the cultivation and selection of ampicillin-resistant E. coli strains. The LB base supports general bacterial growth, and 100 µg/mL ampicillin sodium salt provides selective pressure for maintenance of ampicillin-resistance markers.

Suitable for screening and propagation of plasmid-bearing transformants in E. coli, where β-lactam selection is required.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Ampicillin Sodium Salt
  • Selection of Ampicillin-Resistant Transformants: Isolates colonies of Escherichia coli carrying AmpR plasmids after transformation.

  • Plasmid Maintenance and Expansion: Maintains selective pressure for AmpR plasmids during streaking, colony picking, and overnight growth of E. coli.

  • Colony Screening (Colony PCR/Sanger Prep): Enables picking AmpR colonies of E. coli for rapid insert verification and sequencing.

  • Blue-White Screening: Supports alpha-complementation on Amp plates for cloning when supplemented with X-gal/IPTG in E. coli.

  • Transformation Efficiency and CFU Enumeration: Quantifies viable Amp-resistant E. coli colonies after ligation or Gibson assembly.

  • Protein Expression Workflow Setup: Selects AmpR expression strains (e.g., T7-based) of E. coli prior to induction in liquid culture.

  • Replica Plating and Plasmid Stability Tests: Assesses retention of AmpR plasmids in E. coli under selective pressure.

  • Conjugation/Cloning Workflows: Selects AmpR recipients or donors post-mating or recombineering steps in E. coli.

  • Glycerol Stock Recovery: Recovers and verifies AmpR E. coli strains from frozen stocks before experiments.

  • Educational and Teaching Labs: Demonstrates antibiotic selection and basic cloning using AmpR E. coli on solid media.
