LB Agar Plates with Ampicillin-50 are solid, nutrient-rich agar media that support the growth and selection of ampicillin-resistant E. coli strains. Each plate contains 50 µg/mL ampicillin sodium salt to enable selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Ampicillin Sodium Salt
  • Antibiotic Selection of Transformants: Selects and maintains plasmid-bearing, ampicillin-resistant Escherichia coli clones after transformation.

  • Colony Isolation and Picking: Streaks or spreads to obtain single AmpR colonies of Escherichia coli for downstream cloning and analysis.

  • Colony Screening for Recombinants: Screens transformed Escherichia coli colonies (e.g., PCR or reporter assays) on selective LB–Amp (50 µg/mL) plates.

  • Plasmid Maintenance and Stability Testing: Maintains and assesses stability of AmpR plasmids in Escherichia coli under selection.

  • CFU Enumeration under Selection: Quantifies transformation efficiency by counting colony-forming units of AmpR Escherichia coli.

  • Library Construction and Archiving: Builds and preserves cDNA/genomic or sgRNA libraries carried on AmpR plasmids in Escherichia coli.

  • Replica Plating and Counter-Selection Workflows: Replicates Amp-selected colonies for comparative screening or phenotype verification in Escherichia coli.

  • Reporter and Expression Construct Screening: Evaluates expression or reporter activity from AmpR plasmids in Escherichia coli on selective plates.

  • High-Density Plating on Large Format: Uses 150 mm plates for high-throughput colony arrays, colony lifts, or pooled screens with Amp selection in Escherichia coli.

  • Conjugation/Plasmid Transfer Selection: Selects recipients acquiring AmpR plasmids after mating or transduction in Escherichia coli.
