LB Agar Plates with 1% Glucose and Kanamycin-30 are nutrient agar plates that support the growth and selection of kanamycin-resistant E. coli strains. The addition of 1% glucose enhances metabolic activity and biomass yield, and 30 µg/mL kanamycin enables selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 1.0% Glucose
  • 30µg/mL Kanamycin
  • Selection of Kanamycin-Resistant Transformants: Isolation and propagation of E. coli carrying kanamycin-resistance plasmids at 30 µg/mL.

  • Colony Isolation and Streaking: Streaking for single colonies of KanR strains for downstream cloning and verification.

  • High-Density Library Plating: Arraying large genomic or plasmid libraries on 245×245 mm plates for efficient screening.

  • Replica Plating: Transferring colonies to secondary selective media for phenotype/genotype confirmation.

  • CRISPR and Allelic Exchange Selection: Selecting edited clones using kanamycin resistance markers.

  • Transposon Mutagenesis Screens: Selecting insertion mutants carrying kanamycin cassettes.

  • Conjugation/Mating Selections: Recovering KanR transconjugants after bacterial mating experiments.

  • Expression Control During Cloning: 1% glucose suppresses basal lac-based expression to stabilize toxic or burdening constructs in E. coli.

  • CFU Enumeration of KanR Cells: Quantifying viable counts of antibiotic-resistant populations.

  • Colony Lifts and Screening Assays: Performing membrane lifts for hybridization or activity screens from KanR colonies.
