LB Agar Plates with Ampicillin-100 are a balanced agar medium used for the growth and selection of ampicillin-resistant E. coli strains. The solid agar surface facilitates colony isolation and routine cloning workflows.

These plates contain 100 µg/mL ampicillin sodium salt to enable selection of plasmid-bearing transformants in E. coli cultures.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/ml Ampicillin
  • Antibiotic Selection of Transformants: Plate transformed E. coli on LB-Amp (100 µg/ml) to select colonies carrying bla-encoded, ampicillin-resistant plasmids.

  • Cloning and Plasmid Maintenance: Grow and maintain plasmid-bearing E. coli colonies prior to miniprep or downstream workflows.

  • Colony Screening and PCR: Pick antibiotic-selected colonies for colony PCR, restriction analysis, or sequencing confirmation.

  • Streaking for Single Colonies: Streak mixed or liquid cultures to isolate single ampicillin-resistant colonies.

  • Transformation Efficiency and CFU Enumeration: Quantify transformation performance by counting colony-forming units on selective plates.

  • Plasmid Library Construction and Screening: Maintain and screen combinatorial or genomic libraries under ampicillin selection in E. coli.

  • Pre-culture Selection for Protein Expression: Select expression strains harboring AmpR plasmids before induction in liquid media.

  • Replica Plating and Stability Testing: Assess plasmid retention, segregation stability, or antibiotic resistance profiles by replica plating.

  • Conjugation/Mating Selection: Select transconjugants when transferred plasmids confer ampicillin resistance in E. coli.

  • Stock Preparation: Pick verified colonies from selective plates to establish glycerol stocks for long-term storage.
