LB Agar Plates with Carbenicillin-100 and Chloramphenicol-34 are LB agar media supplemented with 100 µg/mL carbenicillin disodium salt and 34 µg/mL chloramphenicol for growth and dual selection of resistant E. coli strains.

These plates are used for maintenance and selection of plasmid-bearing transformants carrying beta-lactam and chloramphenicol resistance markers, supporting routine cloning, expression, and co-transformation workflows in E. coli.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin Disodium Salt
  • 34µg/mL Chloramphenicol
  • Dual-Plasmid Selection in Escherichia coli: Selects co-transformants carrying AmpR (carbenicillin 100 µg/mL) and CamR (chloramphenicol 34 µg/mL) on LB agar.

  • Transformation and Cloning Workflows: Isolates recombinant E. coli colonies after ligation/Gibson assembly requiring simultaneous AmpR and CamR selection.

  • Maintenance of Co-Expressed Systems: Sustains dual plasmid expression constructs in E. coli for protein production under combined antibiotic pressure.

  • BAC/Plasmid Co-Maintenance: Supports chloramphenicol-resistant BACs with an AmpR helper or expression plasmid in E. coli.

  • CRISPR/Cas Workflows: Selects E. coli cells harboring separate Cas and guide/donor plasmids marked with AmpR and CamR.

  • Library Screening and Colony Picking: Enables high-throughput picking of dual-resistant E. coli colonies for downstream colony PCR and sequencing.

  • Plasmid Stability Assays: Evaluates retention of AmpR- and CamR-marked plasmids in E. coli under dual-antibiotic selection.

  • Conjugation/Mating Selection: Recovers E. coli transconjugants that acquire both resistance markers after transfer events.

  • Reporter and Pathway Engineering: Maintains multi-component metabolic or reporter circuits split across AmpR and CamR plasmids in E. coli.

  • Quality Control of Resistance Cassettes: Verifies correct insertion/expression of beta-lactamase and chloramphenicol acetyltransferase markers in E. coli.
