LB Agar Plates with Carbenicillin-100 are an agar medium that supports the growth and selection of carbenicillin-resistant E. coli strains. The inclusion of 100 µg/mL carbenicillin disodium salt enables effective selection of plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin Disodium Salt

  • Selection of Transformants: Isolates carbenicillin-resistant E. coli carrying bla/β-lactamase plasmids after heat-shock or electroporation.

  • Colony Isolation and Streaking: Streaks for single colonies to obtain clonal carbenicillin-resistant E. coli populations.

  • Plasmid Maintenance and Propagation: Maintains selection pressure to prevent plasmid loss in E. coli cultures.

  • Library and CFU Titration: Quantifies transformation efficiency and library diversity by counting resistant colonies.

  • Blue-White Screening (with supplements): Enables α-complementation screening in E. coli when supplemented with X-gal/IPTG.

  • Colony PCR and Screening: Provides isolated colonies for downstream PCR/verification of inserts from resistant E. coli.

  • Conjugation/Mating Selection: Selects recipients/donors acquiring carbenicillin-resistant plasmids in E. coli matings.

  • Short-Term Strain Storage: Preserves carbenicillin-resistant E. coli strains on plates for routine lab use.
