LB Agar Plates with Kanamycin-25 are a nutritionally balanced solid medium for cultivation and selection of kanamycin-resistant E. coli and related gram-negative bacteria used in molecular cloning. The LB base provides peptides, amino acids, vitamins, and sodium chloride, and agar supplies a firm surface for colony isolation.

The inclusion of 25 µg/mL kanamycin sulfate enables selection and maintenance of cells harboring kanamycin resistance markers, permitting growth of transformants while suppressing non-resistant populations.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 25µg/mL Kanamycin Sulfate
  • Selection of Kanamycin-Resistant Transformants: Isolation of recombinant colonies after transformation of Escherichia coli or other kan-resistant hosts using LB (1.0% Tryptone, 0.5% Yeast Extract, 1.0% NaCl, 1.5% Agar) with 25 µg/mL kanamycin.

  • Plasmid Maintenance and Propagation: Stable selection and passage of plasmids carrying kanamycin resistance cassettes in strains such as Escherichia coli, Agrobacterium tumefaciens, and Pseudomonas putida.

  • Colony Screening and Picking: Growth of individual colonies for downstream colony PCR, restriction analysis, or sequencing under kanamycin selection.

  • Library Construction and Screening: Selection of genomic, cDNA, or mutagenesis libraries cloned into kanamycin-resistant vectors.

  • Protein Expression Strain Selection: Maintenance of expression hosts (e.g., T7-based systems) harboring kanamycin-selectable plasmids prior to induction.
