LB Agar Plates with Carbenicillin-100 are a nutritionally balanced agar medium that supports the growth and selection of carbenicillin-resistant E. coli strains. 100 µg/mL carbenicillin disodium salt provides selection for plasmid-bearing transformants.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin Disodium Salt
  • Antibiotic Selection of bla+ Clones: Selects and maintains Escherichia coli harboring ampicillin/carbenicillin-resistance plasmids (100 µg/mL carbenicillin) on LB agar (1.0% tryptone, 0.5% yeast extract, 1.0% NaCl, 1.5% agar).

  • Cloning and Transformation Screening: Recovery and screening of transformants carrying carbenicillin-selectable plasmids after heat-shock or electroporation.

  • Streaking for Isolation: Single-colony purification of recombinant E. coli strains to ensure clonal populations.

  • Colony PCR and Insert Verification: Picking antibiotic-selected colonies for colony PCR, restriction analysis, or sequencing.

  • CFU Enumeration Under Selection: Quantifying transformation efficiency or viable counts of carbenicillin-resistant cells.

  • Blue-White Screening (With Supplements): α-complementation screening when supplemented with X-Gal/IPTG on carbenicillin plates.

  • Pre-culture Starter Colony Selection: Picking verified colonies to start liquid cultures for minipreps, midipreps, or protein expression.

  • Reduced Satellite Colonies vs Ampicillin: Using carbenicillin to minimize satellite colony formation around β-lactamase+ colonies.

  • High-Throughput Colony Handling: Rectangular 86×128 mm format suited for robotics, replica plating, and automated colony picking.

  • Conjugation/Strain Construction Selection: Selecting recipients/donors carrying bla-marked plasmids in genetic crosses.
