LB Agar Plates with Ampicillin-50 are a nutrient agar medium designed for cultivation and selection of ampicillin-resistant E. coli strains. The formulation is based on Luria-Bertani (LB) agar, providing a general-purpose nutrient base for routine cloning and strain maintenance.

These plates contain 50 µg/mL ampicillin sodium salt to enable selection of plasmid-bearing transformants. Suitable for standard molecular biology workflows involving antibiotic selection in E. coli.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 50µg/mL Ampicillin
  • Plasmid Selection and Maintenance: Selects and maintains AmpR plasmid-bearing Escherichia coli using 50 µg/mL ampicillin on LB agar (1.0% Tryptone, 0.5% Yeast Extract, 1.0% NaCl, 1.5% Agar).

  • Transformation Screening: Recovers and screens heat-shock or electroporation transformants of Escherichia coli for ampicillin resistance.

  • Colony Isolation and Purification: Streaks single colonies to purity prior to liquid culture or glycerol stock preparation.

  • Colony PCR: Provides colonies as DNA templates to verify inserts or vector presence in AmpR clones.

  • Blue-White Screening (with X-Gal/IPTG): Enables α-complementation-based screening of recombinant clones on Amp plates when supplemented.

  • Expression Construct Screening: Screens recombinant Escherichia coli strains harboring AmpR expression vectors before induction in liquid culture.

  • Library Cloning and Subcloning: Selects ampicillin-resistant colonies during construction and refinement of DNA libraries.

  • CFU Enumeration: Quantifies viable counts of AmpR Escherichia coli following transformation or outgrowth.

  • Antibiotic Resistance Verification: Confirms functional AmpR markers and assesses loss/retention of plasmids under selection.

  • Bacterial Conjugation/Mating Selection: Selects transconjugants carrying AmpR plasmids on LB agar with ampicillin.
