LB Agar Plates with Chloramphenicol-30 is a nutrient agar medium that supports the growth and selection of chloramphenicol-resistant E. coli strains. The inclusion of 30 µg/mL chloramphenicol enables selection of plasmid-bearing transformants and maintenance of chloramphenicol-selectable vectors.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 30µg/mL Chloramphenicol
  • Antibiotic Selection of Transformants: Selects colonies carrying chloramphenicol-resistance plasmids in E. coli at 30 µg/mL.

  • Plasmid Maintenance and Propagation: Maintains CmR cloning/expression vectors (e.g., pACYC-derived) in E. coli strains.

  • Post-Cloning Screening: Screens recombinant colonies after ligation, Gibson, or Golden Gate assembly into CmR vectors in E. coli.

  • Colony Isolation and CFU Enumeration: Streaks, isolates single colonies, and performs CFU counts under chloramphenicol selection.

  • Library Plating: Plates genomic, cDNA, or mutant libraries constructed on chloramphenicol-resistant backbones in E. coli.

  • Multi-Plasmid Co-Selection: Supports co-transformation with distinct antibiotic markers including chloramphenicol in E. coli.

  • BAC/PAC/Fosmid Work: Selects and maintains large-insert constructs with chloramphenicol markers in E. coli.

  • Replica Plating and Stability Assays: Assesses plasmid retention and strain stability under chloramphenicol pressure in E. coli.

  • Strain or Vector Verification: Confirms chloramphenicol resistance phenotypes in engineered E. coli strains.

  • Glycerol Stock Preparation: Picks verified chloramphenicol-resistant colonies for long-term storage.
