LB (Lysogeny Broth) Agar Plates with Kanamycin-25 and X-Gal are an agar medium that supports the growth and selection of kanamycin-resistant E. coli strains. 25 µg/mL kanamycin sulfate enables selection of plasmid-bearing transformants, and 60 µg/mL X-Gal supports colorimetric screening of lacZ α-complementation when used with IPTG.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 25µg/mL Kanamycin Sulfate
  • 60µg/mL X-Gal

  • Selection of E. coli transformants (kanR): Isolates colonies carrying kanamycin-resistant plasmids at 25 µg/mL.

  • Blue-white screening (lacZα): Differentiates recombinant (white) from non-recombinant (blue) colonies with 60 µg/mL X-Gal.

  • Colony picking and archiving: Enables reliable recovery of single recombinant clones for downstream work.

  • Colony PCR screening: Rapid insert verification directly from white colonies.

  • Plasmid maintenance and propagation: Stable culturing of kanamycin-selectable vectors in E. coli.

  • Library screening: cDNA/genomic library clone discrimination using kanR and lacZα markers.

  • CRISPR/Cas and genome editing workflows: Selection of kanR editing plasmids in E. coli.

  • Expression construct verification: Pre-expression screening of inserts and reading frames in cloning strains.

  • Transformation efficiency assays: Quantifies competent cell performance using kanR plasmids.
