LB (Lysogeny Broth) Agar with Carbenicillin-100, 60 µg/mL X‑Gal, and 0.3 mM IPTG is a nutrient medium that supports growth and selection of carbenicillin-resistant E. coli strains. Inclusion of 100 µg/mL carbenicillin disodium salt permits selection of plasmid-bearing transformants.

The combination of 60 µg/mL X‑Gal and 0.3 mM IPTG supports blue–white screening via lacZ α‑complementation in appropriate host strains, enabling colony color differentiation during cloning workflows.

Blue-White Screening: Differentiates recombinant from non-recombinant colonies in Escherichia coli via IPTG-induced lacZ α-complementation and X-Gal colorimetric readout.
Antibiotic Selection: Selects for plasmids encoding β-lactam resistance using 100 µg/mL carbenicillin in Escherichia coli.
Plasmid Maintenance: Maintains cloned constructs in Escherichia coli on LB agar (1.0% tryptone, 0.5% yeast extract, 1.0% NaCl, 1.5% agar).
Colony Picking for Downstream Workflows: Enables isolation of individual colonies for miniprep, Sanger sequencing, colony PCR, or glycerol stock preparation.
Transformation Efficiency Assessment: Quantifies CFUs of transformants on selective IPTG/X-Gal plates in Escherichia coli.
Library Screening: Screens combinatorial or mutant libraries by color and antibiotic resistance in Escherichia coli.
Cloning Verification: Confirms insertional inactivation of lacZα and construct assembly (e.g., Gibson or Golden Gate) in Escherichia coli.
Strain/Vector QC: Validates lacZ α-complementation capability and vector backbones using IPTG (0.3 mM) and X-Gal (60 µg/mL) in Escherichia coli.
