LB Agar Plates with Carbenicillin-100 and Kanamycin-25 are solid nutrient media for cultivation and dual-antibiotic selection of E. coli strains. The inclusion of 100 µg/mL carbenicillin and 25 µg/mL kanamycin enables selection of transformants carrying beta-lactam and aminoglycoside resistance markers during routine streaking and colony isolation.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 100µg/mL Carbenicillin
  • 25µg/mL Kanamycin
  • Dual-antibiotic selection of transformants: Isolating recombinant Escherichia coli carrying plasmids with beta-lactam (bla) and kanamycin (nptII/aph) resistance markers.

  • Co-transformation and plasmid co-maintenance: Selecting Escherichia coli clones harboring two compatible plasmids requiring carbenicillin and kanamycin.

  • Cloning and assembly screening: Identifying correct colonies after Gibson, Golden Gate, or ligation under dual selection pressure.
