LB Agar Plates with IPTG and X-Gal are a nutritionally balanced agar medium for cultivation of E. coli and blue/white screening via lacZ α-complementation. IPTG provides non-metabolizable induction of β-galactosidase, and X-Gal functions as a chromogenic substrate to differentiate colonies based on β-galactosidase activity.

  • 1.0% Tryptone
  • 0.5% Yeast Extract
  • 1.0% Sodium Chloride
  • 1.5% Agar
  • 60µg/mL X-Gal
  • 0.1mM Isopropyl-B-D-thiogalactopyranoside(IPTG)
  • Blue-White Screening of Recombinant Clones: α-complementation-based selection of inserts in lacZα plasmids using E. coli.

  • Bacterial Transformation Screening: Rapid identification of transformants carrying inserts after chemical or electroporation methods in E. coli.

  • Colony Picking and Clone Isolation: Distinguish blue (non-recombinant) from white (recombinant) colonies for downstream plasmid prep and sequencing.

  • Library Construction and Screening: cDNA/genomic library screening via lacZα disruption to enrich for insert-containing clones in E. coli.

  • TA/TOPO Cloning Workflows: Visual selection of recombinant inserts in lacZα-based TA or TOPO vectors in E. coli.

  • Phage Plaque Color Screening: Blue/white plaque discrimination for recombinant M13 or λ-based lacZα phage systems.
